Semen analysis is done to study the fertility status of a male.
Semen should be collected in a clean dry container having no detergent.
Is to be collected by masturbation or by coitus interruptus.
The following features are to be noted:
2. Colour: White in colour.
3. Consistency: Turbid & Viscid.
4. Total spermatozoa: 20-150 million/ml.
Laboratory Examination:
1. Amount: Nornal- at least 0.5 ml in one ejaculation.
5. Method: Three parts of 5% formalin & one part of saturated gention violet in alcohol is used to stain spermatozoa. Distilled water is tinged with this stain.
Semen is diluted 1 in 10 with this water in WBC pipette. A counting chamber is filled. The number of spermatozoae in 5 small squares (as done in RBC count) are counted. Total spermatozoae is obtained by multiplying the number by 10,00,000.
Motility of spermatozoa: Normal: 40-100%.
Morphology: Normal: 75-100%.
Method: A cover slip preparation is made as done for the motility test of bacteria and examined under high power of microscope.
Method: A film is prepared on a clean glass slide and stained with the stain mentioned under total count. Morphology of head neck and tail are recorded.
Opinion:
nNormozoospermia: If the sperm count is more than 20 million/ml.
nOligospermia: If the sperm count is less than 20 million/ml and have abnormal morphology.
nAzoospermia: If spermatozoa is absent in the semen.
nHyperzoospermia: If the sperm count is more than 150 million/ml.
